Abstract
ENGLISH ABSTRACT: In this study the possible use of acid-treated, naked bacteria as oral vaccines was investigated using TNP-NB conjugates and LPS/NB complexes as model antigens. A novel solid-phase enzyme-linked immunospot (ELISPOT) assay for the detection of specific antibody secreting cells was developed and used to determine the amount of specific antibody secreting cells in the spleens and smallintestines of mice im,unised with TNP-NB subjugates. It was found that large numbers of anti-TNP antibody secreting cells were produced in the wall of the small intestine in response to oral immunisation with TNP-NB conjugates.
AFRIKAANSE OPSOMMING: In hierdie studie is die moonlike gebruik van suur-behandelde, naakte bakterieë as mondeliks toegediende entstowwe bestuur deur gerbuik te maak van TNP-NB konjugate en LPS/NB komplekse as model antigene. 'n Nuwe soliede fase ensiem gekoppelde immunospot (ELISPOT) essai is ontwikkel om die hoeveelheid spesifieke antiligaam-produserende selle in die milt en dunderm van muise, wat met TNP-NB konjugate geïmuniseer is, vas te stel. Groot hoeveelhede selle wat spesifieke anti-TNP antiliggame produseer, is gevind in die wand van die muis dunderm, wat daarop dui dat daar 'n spesifieke immuunrespons plaasvind na mondelikse toediening van TNP-NB konjugate.
