species has been carried out by using epifluorescence and
confocal laser scanning microscopy. The analysis of the
autofluorescence emitted by leaf tissues, represents a rapid,
easy and non-disruptive method to determine the basic
histological structure of both photosynthetic and non-photosynthetic
tissues. The present study allowed us to classify
the plants analyzed into several categories on the basis of
their structure, as well as to suggest a mixed C4 / CAM nature
for some of them.
The results obtained here indicate that confocal microscopy,
in addition to significantly improving the resolution over
standard epifluorescence, may allow the development of
new applications such as the discrimination of the specific
distribution of photosystems I and II in C4 plants, or the
quantification of the activity of the different components of
the photosynthetic apparatus.
The use of these Cell Biology tools, will allow the design of
new experiments in order to improve the knowledge regarding
the physiology of these plants, by analyzing the autofluorescence
emitted by their photosynthetic apparatus under
several environmental conditions. [ES]: Se ha realizado un estudio histológico de hojas de diversas plantas C4 y CAM mediante microscopía de epifluorescencia y microscopía de barrido láser confocal. El análisis de la autofluorescencia emitida por los tejidos de la hoja constituye un método rápido, sencillo y no disruptivo para establecer la organización básica de los tejidos fotosintéticos y no fotosintéticos. Dicho estudio ha permitido clasificar las plantas analizadas en varias categorías estructurales, además de sugerir un carácter mixto C4 / CAM para algunas de ellas. Los resultados obtenidos indican que la microscopía confocal, aparte de mejorar ostensiblemente la resolución respecto a la epifluorescencia estándar, probablemente permita desarrollar nuevas aplicaciones, como discriminar la distribución específica de los fotosistemas I y II en plantas C4, o cuantificar la actividad de los diferentes componentes del aparato fotosintético. La utilización de estas herramientas de Biología Celular permitirá, gracias a su facilidad de uso, diseñar experimentos destinados a ampliar el conocimiento de la fisiología de estas plantas, analizando la autofluorescencia específica emitida por su aparato fotosintético en distintas condiciones ambientales. |
