Abstract: Root canal infections are polymicrobial Coaggregation between bacteria in the root canal play a potential role in endodontic infection The aim of this study was to investigate the antibacterial efficacy of 3 per cent sodium hypochlorite and 2 per cent chlorhexidine pretreated with coaggregation inhibitors against dual species biofilm of E faecalis and F nucleatum 375 Dentine samples from human uniradicular teethwere taken 45 samples were inoculated with mono species biofilm of E faecalis and fusobacterium nucleatum and dual species to check the depth of penetration and biovolume 240 samples n equal to 15 were pretreated with 100 mM of coaggregation inhibitors lactose N Acetyl Dgalactosamine LArginine LLysine and disinfection was done with 3percent sodium hypochlorite NaOCl and 2 per cent chlorhexidine chx 60 samples n 15 were treated with 3 per cent NaOCl and 2 per cent CHX 30 samples were used as positive control with dual species biofilm The remaining biovolume and bacterial quantification was analyzed using confocal microscopy and molecular probe assay rt PCR Taqman probe A significant decrease in the percentage of biovolume and bacterial number after pretreatment with 100 mMol of LArginine followed by NAcetyl galactosamine before 3 percent NaOCl disinfection compared to 3 percent NaOCl without pretreatment and positive control no treatment p less than 0 point 05 Coaggregation inhibitors enhanced the antimicrobial activity of 3 percent sodium hypochlorite and 2 percent Chlorhexidine LArginine pretreatment before disinfection with 3 percent NaOCl showed the least percentage of bacterial volume and number of dual species of Efaecalis and Fnucleatum newline
