Abstract: Background: Gandhaka pushapa is a folklore medicinal preparation. Preparation time contents were going blooms like flower and hence named as Gandhaka pushpa. This is being used widely by Ayurved physicians for the purpose of rakta shodhaka, vrana ropaka, in the management of kustha. There is a paucity of safety and efficacy of Gandhakapushpa prepared by folklore method. It is proposed that, if the preparation is prepared by following standard Ayurvedic preparation methods and studied for safety and toxicity, and then the preparation may be used widely. Objectives: Preparation of Gandhakapushpa according to Ayurvedic method Investigation of Antimicrobial activity of Gandhakapushpa prepared by Ayurvedic and folk methods To evaluate safety of Gandhakaushpa prepared by Ayurvedic and folklore methods in wistar rats Methodology: Gandhaka and Erandabeeja are selected according to Grahya lakshana, and Shodhana and murchana sanskara are conducted according to Ayurvedic methods. Antimicrobial activity study (Cup plate method and MIC method) are conducted for both Gandhakapushpa samples. One day acute and 90 days chronic studies are conducted on Wistar female albino rats by using both the samples. Results: 1) In quantitative (MIC) method Gandhakapushpa prepared by Ayurvedic method is effective against fungi and gram negative organisms i.e. 2mg/ml and 4.01 mg/ml concentrations respectively. Folklore prepared Gandhakapushpa shows effective against fungi (Candida albicans) i.e. 0.25mg/ml concentration. 2) LD50 of Gandhakapushpa prepared by Ayurvedic method and folklore prepared Gandhakapushpa is found to be more than 5000 mg/kg bd wt. 3) In single dose acute study Gandhakapushpa prepared by Ayurvedic method at dose 500 mg/kg bd wt showed no toxic effects on vital organs and hematological parameters whereas folklore prepared Gandhakapushpa shows toxic effects on vital organs and hematological and serum biomarkers. 4) In chronic study Gandhakapushpa prepared by Ayurvedic method in the dose 1000 mg/kg bd.wt treated animals shows less toxic effects then compare to folklore prepared Gandhakapushpa which shows severe toxic effect on the vital organs and hematological as well as serum biomarkers in the dose 500mg/ kg bd wt. Conclusion: Gandhakapushpa of both preparation methods exhibit antimicrobial property. However Gandhakapushpa prepared by folklore practice is found toxic. Hence Gandhakapushpa prepared by Ayurvedic method may be used externally for the management of skin infections caused by Kleb siella, Candida albicans and Aspergillus niger organisms. newline
