Using truncated and mutated NMDA receptor subunits expressed in heterologous cells, we found that the M3 domains of both NR1 and NR2 subunits contain at least two structural determinants that regulate the number of surface functional NMDA receptors. These structural determinants are neither critical for the interaction between the NR1 and NR2 subunits nor for the formation of the functional receptors, but rather they regulate the release of the receptors from the ER
