In this work, the analytical characteristics of the enzyme sensors of glucose with an optical oxygen transducer embedded in organic-inorganic polymer ORMOCER ® (GOR) were studied. The robust sensor was proposed for the online monitoring of biotechnological processes. The scope of the thickness, ratio of the components of the optically sensitive layers and glucose oxidase concentration in the sensitive layers with particular structure was limited by the mechanical stability of the layers and the sensitivity of the electro-optical instrumentation. These changes resulted in GOR with the sensitivity from 0.001-0.452 µs • l mmol -1 and linear dynamic range (LDR) from 0-75.0 mM. The fastest response of the prepared biosensors was 1.6 min. A further extension of the range of analytical characteristics was realized by changing the concentration of dissolved oxygen (DO) in the measured solutions. The LDR was increased more than ten times by increasing the DO from 5% to 100%. In order to estimate the analytical characteristics of the GOR sensor in the parametrical space, a mathematical model of the reaction-transport phenomena inside the optically sensitive layers was developed. This model, consisting of two reaction-diffusion equations for the glucose and oxygen substrates, was solved numerically. The dependence of the dynamics of the glucose and oxygen concentration in the sensitive layers on two dimensionless parameters was studied. One of them represents the ratio of the characteristic rates of the diffusion transport and enzyme reaction. This parameter couples the effects of the thickness of the layers and enzyme activity. The other parameter depends on the diffusivities of glucose and oxygen in the sensitive layers. The theoretical findings corresponded to the experimental data. Thus, the results of the mathematical modelling may serve as guidelines in the design of GOR and other enzyme sensors with optical oxygen transducers.
