Une lésion persistante du foie peut provoquer une fibrose hépatique (FH), une cirrhose et un carcinome hépatocellulaire. La compréhension de la pathogenèse moléculaire de la FH pourrait conduire à des nouveaux traitements pour ces maladies. Le suppresseur de la signalisation de cytokine 1 (SOCS1) régule plusieurs cytokines et facteurs de croissance impliqués dans la pathogénèse de la FH. Ici, nous avons étudié la fonction anti-fibrogénique de SOCS1 dans les hépatocytes et les macrophages.
Des souris dépourvues de SOCS1 dans les hépatocytes (Socs1fl/fl AlbCre; Socs1Hep) ou des macrophages (Socsfl/fl LysMCre; Socs1M) ont été générées et nous avons évalué la réponse fibro-hépatique au tétrachlorure de carbone (CCl4). Les lésions hépatiques et la fibrose ont été évaluées par les taux d’Alanine Aminotransférase (ALT), l'histopathologie et le taux d’hydroxyproline dans le foie. Les changements dans l'expression des enzymes de la matrice extracellulaire et du remodelage, des cytokines et des chimiokines ont été évalués par qRT-PCR. Les souris Socs1fl/fl AlbCre et Socsfl/fl LysMCre ont présenté une FH sévère caractérisée par une augmentation en dépôt de collagène, de la teneur en hydroxyproline, de l'accumulation de myofibroblastes et de l'expression des gènes Acta2 et Coa1a1. Le traitement par CCl4 a causé des dommages significatifs aux hépatocytes chez les souris Socs1fl/fl AlbCre mais pas chez les souris Socsfl/fl LysMCre. Les deux souris ont montré une expression réduite de Mmp2 et une augmentation de l'expression de Timp1. L'induction de l'Il6, Tnfa et Tgfb n'était pas significativement altérée par une déficience en SOCS1 dans les hépatocytes ou dans les macrophages, ce qui diminuait l’INF et augmentait l'expression de Pdgfb. Les foies Socs1fl/fl AlbCre et Socsfl/fl LysMCre présentaient une augmentation de l'infiltration des cellules mononucléaires associée à une expression élevée de Ccl2, tandis que les foies Socsfl/fl LysMCre présentaient également une expression accrue de Cx3cr1.
SOCS1 est donc un régulateur clé de la FH. Il exerce ses fonctions anti-fibrotiques dans les hépatocytes et les macrophages. Alors que le SOCS1 dans les hépatocytes atténue la mort cellulaire, dans les cellules myéloïdes il contrôle la cascade inflammatoire conduisant à la FH.
Abstract : Background & Aim: Persistent liver injury causes liver fibrosis (LF), cirrhosis and hepatocellular carcinoma (HCC). Understanding of the molecular pathogenesis of LF could lead to new treatments for these diseases. Suppressor of Cytokine Signaling 1 (SOCS1), which regulates several cytokines and growth factors implicated in LF, attenuates LF. Here, we investigated the anti-fibrogenic function of SOCS1 in hepatocytes and macrophages.
Methods: We have generated mice lacking SOCS1 in hepatocytes (Socs1fl/flAlbCre) or macrophages (Socs1fl/flLysMCre) and evaluated hepatic fibrogenic response to carbon tetrachloride (CCl4). Liver damage and fibrosis were evaluated by serum Alanine Amino Transferase(ALT) levels, histopathology and hydroxyproline content. Changes in the expression of extracellular matrix and remodeling enzymes, cytokines and chemokines were assessed by qRT-PCR.
Results: Socs1fl/flAlbCre and Socs1fl/flLysMCre mice showed severe LF characterized by increased collagen deposition, hydroxyproline content, myofibroblast accumulation and expression of Acta2 and Coa1a1 genes. CCl4 treatment caused significant damage to hepatocytes in Socs1fl/flAlbCre mice but not in Socs1fl/flLysMCre mice. Both mice showed reduced expression of Mmp2 and increased expression of Timp1. Il6, Tnfa and Tgfb induction were not significantly altered by SOCS1 deficiency in hepatocytes or macrophages, which diminished INF and augmented Pdgfb expression. Both Socs1fl/flAlbCre and Socs1fl/flLysMCre livers showed increased mononuclear cell infiltration accopmanied by elevated Ccl2 expression, whereas Socs1fl/flLysMCre livers also showed increased Cx3cr1 expression.
