De neurotransmitter dopamine (DA) is betrokken bij hersenfuncties als beweging, cognitie en emotioneel- en beloningsgerelateerd gedrag. Het afsterven van DA-neuronen in de substantia nigra in de hersenen veroorzaakt de ziekte van Parkinson. DA-stoornissen in andere hersengebieden worden in verband gebracht met schizofrenie, verslaving, obsessief-compulsieve stoornissen, het syndroom van Gilles de la Tourette en ADHD. Pepijn van den Munckhof onderzocht de aphakia-muis, die van nature geen Pitx3-gen heeft. Hierdoor ontstaat een groot tekort aan dopamineneuronen in de substantia nigra. De verschijnselen zijn vergelijkbaar met die van Parkinsonpatiënten; de aphakiamuis vormt hier dan ook een goed diermodel voor. In patiënten worden meer veranderingen gezien, maar toch lijkt het Pitx30-gen van cruciaal belang bij overleving van DA-neuronen in het de substantia nigra.
