Estresses abióticos na germinação de sementes e no crescimento de mudas de espécies e híbridos de Eucalyptus

Resumo

RESUMO- O gênero Eucalyptus é um dos mais importantes para a silvicultura brasileira, sendo fonte de matéria-prima para diversos produtos. As alterações climáticas que vem ocorrendo nos últimos anos tem influenciado negativamente a produtividade florestal, principalmente, devido ao aumento da temperatura e mudanças na intensidade e duração das secas. A seleção de genótipos mais produtivos e tolerantes para as diversas condições é um dos objetivos do melhoramento genético de espécies de eucalipto, contudo, por ser uma planta perene, leva - se muito tempo para se obter resultados satisfatórios. Buscar meios mais rápidos para seleção de materiais, é importante para agilizar o processo. Com isso o presente trabalho teve como objetivo avaliar o efeito da temperatura, do déficit hídrico e estresse salino na germinação de sementes e avaliar o desempenho de mudas sob déficit hídrico de cinco espécies e dois híbridos de eucalipto. A pesquisa foi dividida em quatro experimentos, os três primeiros foram testes de germinação, feitos em laboratório e a avaliação de mudas, em vasos em casa de vegetação. As espécies e híbridos (genótipos) de eucalipto utilizadas foram, Eucalyptus camaldulensis, E. brassiana, E. grandis, E. saligna, E. urophylla, E. grandis x E. camaldulensis e E. urophylla x E. grandis. O primeiro experimento consistiu de um delineamento em blocos casualizados em esquema fatorial 5x7, sendo cinco temperaturas (20, 25, 30, 35 e 40°C) e os sete genótipos. No segundo experimento as sementes foram submetidas à germinação em temperatura ótima (25º C) e supra-ótima (35º C) e nos potenciais hídricos de 0(água); -0,05; -0,10; -0,15; -0,20; -0,25; -0,30; -0,40; -0,60; -0,80 e -1,00 MPa, obtidos com polietilenoglicol (PEG 6000). O experimento foi conduzido em delineamento de blocos casualizados, em arranjo fatorial 11x7x2 (11 potenciais osmóticos x 7 genótipos x 2 temperaturas). O terceiro experimento constou do mesmo arranjo do anterior, envolvendo, porém, o estresse salino, obtendo os mesmos potenciais hídricos, com diluições de NaCl em água. O último experimento foi em vasos em casa de vegetação, utilizando o delineamento em blocos casualizados em esquema fatorial 3x7, sendo três regimes hídricos (60, 40 e 20% da capacidade de campo) e os sete genótipos de eucalipto. As variáveis analisadas nos experimentos em laboratório, foram germinação, índice de velocidade de germinação, coeficiente de germinação, tempo médio de germinação e comprimento de parte aérea e de raiz. Em casa de vegetação foram avaliadas características de crescimento (diâmetro do coleto, altura de plantas, número de folhas, área foliar, massa fresca de parte aérea e de raiz), fisiológicas (taxa de assimilação líquida, transpiração, condutância estomática, potencial hídrico foliar) e bioquímicas (conteúdo de carboidratos solúveis e totais, conteúdo de clorofila a e b e de carotenoides) e morfológicas (morfologia e número de estômatos da folha). Os dados obtidos foram submetidos à análise de variância e comparação de médias pelo teste de Tukey, a 5% de probabilidade para comparação entre os genótipos e para os efeitos de potenciais hídricos foi aplicado uma análise de regressão. Os genótipos apresentaram comportamento diferenciado frente às temperaturas estudadas. Até 30 ºC não houve diferença na germinação dos genótipos, diferindo somente nas temperaturas mais elevadas, 35 e 40º C. O E. camaldulensis e E. brassiana apresentaram maiores porcentagens de germinação que os demais. No segundo experimento houve redução na porcentagem e no índice de velocidade de germinação das sementes com a diminuição do potencial hídrico e, em geral, com o aumento da temperatura. E. camaldulensis e E. brassiana se sobressaíram com maior capacidade de germinação no potencial hídrico de -0,40 MPa, comparativo aos outros genótipos. O mesmo também ocorreu sob estresse salino, porém, houve maior capacidade de germinação em maiores concentrações de NaCl, chegando a obter germinação em potenciais de -0,80 MPa. Em casa de vegetação os genótipos apresentaram diferenças nas características avaliadas que resultaram em maior ou menor grau de tolerância ao déficit hídrico. De modo geral, E. camaldulensis e E. brassiana também se apresentaram superiores, principalmente quanto ao desempenho fisiológico, quando comparado aos demais genótipos. Em conjunto, os resultados indicam possível relação entre as fases de germinação e desempenho inicial das mudas, quanto às respostas aos estresses abióticos testados. A seleção de genótipos tolerantes aos estresses abióticos na germinação ainda é precipitada, porém, através deste, viu-se que é possível indicar o local onde as plantas apresentarão melhor desempenho.
ABSTRACT- The genus Eucalyptus is one of the most important for Brazilian forestry, being source of raw material for several products. The climate change that has been occurring in recent years has negatively influenced forest productivity, mainly due to the increase in temperature and changes in the intensity and duration of droughts. The selection of more productive and tolerant genotypes for the different conditions is one of the objectives of the genetic improvement of eucalyptus species, however, because it is a perennial plant, it takes a long time to obtain satisfactory results. Finding faster means for material selection is important to streamline the process. The objective of this work was to evaluate the effect of temperature, water deficit and saline stress on seed germination and evaluate the performance of seedlings under water deficit of five species and two eucalyptus hybrids. The research was divided in four experiments, the first three were tests of germination, done in laboratory and in the fourth experiment were evaluated of seedlings, in pots under greenhouse conditions. The eucalyptus species and hybrids (genotypes) used were Eucalyptus camaldulensis, E. brassiana, E. grandis, E. saligna, E. urophylla, E. grandis x E. camaldulensis and E. urophylla x E. grandis. The first experiment consisted of a randomized block design in a 5x7 factorial scheme, with five temperatures (20, 25, 30, 35 and 40 ° C) and the seven genotypes. In the second experiment the seeds were submitted to germination at optimum temperature (25ºC) and supra-optimum (35ºC) and in water potentials of 0 (water); -0.05; -0.10; -0.15; -0.20; -0.25; -0.30; -0.40; -0.60; -0.80 and -1.00 MPa, obtained with polyethylene glycol (PEG 6000). The experiment was conducted in a randomized complete block design, in factorial arrangement 11x7x2 (11 osmotic potentials x 7 genotypes x 2 temperatures). The third experiment consisted of the same arrangement of the previous one, involving, however, saline stress, obtaining the same water potentials, with dilutions of NaCl in water. The last experiment was in pots under greenhouse conditions, using a randomized block design in a 3x7 factorial scheme, with three water regimes (60, 40 and 20% of field capacity) and seven eucalyptus genotypes. The variables analyzed in the laboratory experiments were germination, germination speed index, germination coefficient, mean germination time, and shoot and root length. In greenhouse conditions, were evaluated growth characteristics (shoot diameter, plant height, leaf number, leaf area, fresh shoot and root mass), physiological (net assimilation rate, transpiration, stomatal conductance, leaf water potential) and biochemical characteristics (soluble and total carbohydrate content, chlorophyll a and b and content of carotenoids) and morphological characteristics (morphology and number of leaf stomata). The data were submitted to analysis of variance and comparison of means by Tukey test, a 5% probability for comparison between genotypes and for the effects of water potentials a regression analysis was applied. The genotypes showed a different behavior compared to the temperatures studied. At 30 ºC there was no difference in germination of the genotypes, differing only at the highest temperatures, 35 and 40 º C. The E. camaldulensis and E. brassiana presented higher germination percentages than the others. In the second experiment there was a reduction in seed germination percentage and seed germination rate, with decrease in water potential and, in general, with temperature increase. E. camaldulensis and E. brassiana showed greater germination capacity in the water potential of -0.40 MPa, compared to the other genotypes. The same occurred also under saline stress, however, there was a greater capacity of germination in higher NaCl concentrations, obtaining germination in potentials of -0.80 MPa. In greenhouse the genotypes presented differences in the characteristics evaluated that resulted in a greater or lesser degree of tolerance to the water deficit. In general, E. camaldulensis and E. brassiana were also superior, mainly regarding the physiological performance, when compared to the other genotypes. Together, the results indicate a possible correlation between the germination stages and initial seedling performance, regarding the responses to the abiotic stresses tested. The selection of genotypes tolerant to abiotic stresses in germination is still precipitated, but through this, it was possible to indicate where the plants will perform better.
