Short chain alkyl monoglucosides (octyl and nonyl-β-glucoside) are soluble in aqueous solutions of ammonium sulfate up to 4 M ionic strength. In combination with PEG of molecular weight from 3350 to 10000, two phase transitions can be observed while increasing temperature, with lower and upper two-phase regions separated by a single isotropic solution. The one-phase region narrows on increasing the concentration of polymer and surfactant, and for PEG 8000 and at high ratios of polymer to glucoside, becomes a small channel where solutions are sensitive to shear. In solutions with octyl-β-glucoside the lower two-phase body is shifted to higher concentrations than the one in solutions with nonyl-β-glucoside, while the upper two-phase body does not vary significantly. The implications of these patterns of phase behavior for membrane protein crystallization will be discussed.
