SYTHEIS AND PURIFICATON OF NSP4150-175 THROUGHT THE USE OF SOLID PHASE RESIN
Abstract
NSP4 is the viral-encoded, enterotoxic protein that is associated with the pathogenesis of rotavirus. Currently, the sequence of NSP4 is known for the different strains of rotavirus, but other proteins that are tightly bound to NSP4 in cells are unknown. In order to study NSP4 and the associated protein complexes the C-terminal segment NSP4150-175 was synthesized through the using solid-phase, FMOC chemistry and Rink amide resin. Synthesis was completed using HOBT activated amino acids and piperidine as the deblocking solution. Peptides will be cleaved from the solid-phase resin using tri-fluoroacetic acid with free-radical scavengers. Purification will be accomplished through gel filtration and HPLC reverse-phase chromatography and verified by mass spectrometry.
