Role of AMP-activated protein kinase (AMPK) in the control of skeletal muscle protein synthesis and epithelial cell architecture

(eng) The AMP-activated protein kinase (AMPK) system has been principally considered as a regulator of cellular and systemic energy balance. However, it is now recognized that its role extends beyond energy sensing to the control of cell division, cell polarity and cell migration. In this thesis, a study of the AMPK system on the control of one energy consuming process (protein synthesis) is presented as well as a study of one non-metabolic function of AMPK, namely control of the actin cytoskeleton. The mechanisms of control of skeletal muscle protein synthesis by contraction/AMPK and insulin were first investigated. In incubated rat epitrochlearis skeletal muscles, insulin stimulated protein synthesis. Contraction induced by electrical stimulation on the other hand led to AMPK activation and inhibition of the basal and insulin-stimulated rates of protein synthesis. Electrical stimulation had no effect on basal mammalian target of rapamycin complex 1 (mTORC1) signaling, but did antagonize mTORC1 signaling after stimulation of the pathway by insulin. Phosphorylation of eukaryotic elongation factor-2 (eEF2), a known target of AMPK, increased rapidly upon electrical stimulation, but could not be related to AMPK activation, which occurred more slowly to reach a 3-fold activation after 30 min. Therefore, in electrically stimulated skeletal muscle, the contraction-induced inhibition of protein synthesis could not be attributed to inhibition of mTORC1 signaling but could be due to increased eEF2 phosphorylation, independent of AMPK activation. The non-metabolic role of AMPK in controlling reorganization of the actin cytoskeleton was studied in epithelial Madin-Darby canine kidney (MDCK) cells. The effects of hyperosmolarity and treatment with pharmacological AMPK activators (AICAR and A-769662) on cytoskeletal organization and signaling were investigated. AMPK activation by A-769662 induced peripheral F-actin accumulation, disappearance of focal adhesions and disassembly of stress fibers. In parallel, RhoA was activated, associated with an increase in phosphorylation of the Rho-kinase (ROCK) downstream targets, myosin regulatory light chain (MLC) and cofilin. Accordingly, the A769662-induced increase in MLC and cofilin phosphorylation was prevented by H1152, a ROCK inhibitor. In MDCK cells hyperosmotic shock activated AMPK via the calmodulin-dependent protein kinase kinase-b pathway. Cell shrinkage induced similar changes in cell architecture and increased MLC phosphorylation to those observed with pharmacological AMPK activators. However, AMPK activation due to ATP-depletion by 2-deoxyglucose treatment did not induce cytoskeletal reorganization or increase MLC phosphorylation in these cells. We propose that AMPK participates via the RhoA/ROCK pathway, in the reorganization of the actin cytoskeleton induced by osmotic stress.

(fre) L’AMPK (AMP-activated protein kinase) participe à l’homéostasie énergétique cellulaire et systémique. De plus, son rôle s’étend au-delà du contrôle énergétique et s’applique au contrôle de la division, de la polarité et de la migration cellulaire. Dans ce travail de thèse, nous avons étudié l’implication de l’AMPK dans le contrôle d’un mécanisme consommant de l’énergie à savoir la synthèse protéique, ainsi que dans le contrôle du cytosquelette d’actine. Nous avons analysé les mécanismes de contrôle de la synthèse protéique par la contraction, l’AMPK et l’insuline dans le muscle squelettique epitrochléaire de rat. L’insuline stimule la synthèse protéique tandis que la contraction induite par la stimulation électrique active l’AMPK et inhibe la synthèse protéique basale et celle induite par l’insuline. La stimulation électrique n’a pas d’effets sur le niveau basal de la voie de signalisation de mTORC1 (mammalian target of rapamycin complex 1) mais elle inhibe l’activation de la voie mTORC1 induite par l’insuline. La phosphorylation de eEF2 (eukaryotic elongation factor-2), une cible en aval de l’AMPK, augmente rapidement en stimulation électrique mais ne peut être imputée à l’activation de l’AMPK qui est plus lente. En conclusion, l’inhibition de la synthèse protéique dans les muscles squelettiques soumis à une stimulation électrique ne peut-être imputée à l’inhibition de la voie mTORC1 mais pourrait résulter d’ une augmentation de la phosphorylation de eEF2, indépendante de l’activation de l’AMPK. Le rôle de l’AMPK dans le contrôle de la réorganisation du cytosquelette d’actine a été étudié dans les cellules épithéliales MDCK (Madin-Darby canine kidney). Les effets des activateurs pharmacologiques (AICAR et A-769662) de l’AMPK et de l’hyperosmolarité sur l’organisation et la signalétique du cytosquelette d’actine ont été investigués. L’activation de l’AMPK par l’A-769662 induit une accumulation périphérique de la F-actine, la disparition des adhésions focales et un désassemblage des fibres de stress. En parallèle, RhoA est activé et impliqué dans la phosphorylation des cibles de ROCK (Rho-kinase) qui sont MLC (myosin light chain) et la cofiline. En effet, l’augmentation de la phosphorylation de MLC et de la cofiline est supprimée en présence de H1152, un inhibiteur de ROCK. L’hyperosmolarité produit des changements de l’architecture cellulaire tout à fait similaires à ceux induits par les activateurs pharmacologiques de l’AMPK et elle conduit également à une augmentation de la phosphorylation de MLC et de la cofiline. Dans les cellules MDCK, le choc hyperosmotique active l’AMPK via une voie de signalisation dépendante de la CaMKKβ (calmodulin-dependent protein kinase kinase-β), une AMPK-kinase. En effet, l’activation de l’AMPK ainsi que la phosphorylation de MLC et de la cofiline induits par le choc hypersosmotique sont réduits par le STO-609, un inhibiteur spécifique de la CaMKKβ. Nous en concluons que l’AMPK participe à la réorganisation du cytosquelette d’actine lors du choc hyperosmotique, via l’activation de la voie RhoA/ROCK.
