The waste cooking oils (WCO) are a difficult management waste stream, therefore a significant part of them do not have an appropriate destination. Currently, the production of biodiesel is one of the most promising recovery operations of this waste, in order to be an alternative energy source to fossil fuels. The increasing interest to use biofuels as substitutes for petroleum fuels are: the climate changes, increasing oil prices and security of energy supply.. Typically, biodiesel is produced through the transesterification reaction between an vegetable oil and an alcohol in the presence of a catalyst. The whole process development is closely related to the type of catalyst and strongly depends on him. The main objective of this work was to investigate the activity loss of the enzyme Novozyme® 435 throughout the several using cycles. For this, the transesterification reaction was carried out by two distinct methods: the use of solvent (tert-butanol) and addition of methanol through stages. The virgin enzyme was pre-treated and the operating conditions were: T = 40ºC , mass ratio catalyst:WCO=10%, molar ratio methanol:OAU=4:1, volume ratio of solvent 0.75 v/v (solvent method) and orbital agitation 200 rpm. The highest yield achieved esters content was 96% in the using solvent method and 75% when methanol was added in stages, both attained in the first cycle of the enzyme use. In either method there was a loss of enzyme activity from cycle to cycle, which was more evident in the solvent method. The enzyme regeneration was tested in both methods. It was found that this procedure did not improve the enzyme activity in any of the methods. In other words, the esters contents did not increased in comparison to those value obtained in the previous cycle. More, they decreased. Out of curiosity, it was analyzed some properties of produced biodiesel and it was observed that almost all limits imposed by EN ISO 14214:2003 were not fulfilled. The aims of this thesis have been achieved. In literature there are few studies on biodiesel production via enzymatic catalysis, especially when the raw material is the WCO. The current work was intended to give a contribution in the area of biofuels, which is also a viable alternative for recover a difficult management waste.
