In this work, we focus on purification of positively charged labeled oligosaccharides from an excess of the unreacted label. The signal of the unreacted label is routinely significantly higher, indicating the need to deplete the unreacted label prior to analysis by CE. Purification can lead to higher sensitivity and precision of the analysis due to suppression of interferences from the unreacted label and the possibility to use more concentrated samples. For this purpose, we tested several solid phase extraction (SPE) columns such as ZIC-HILIC, amide, diol, boronate, carbon, etc. phases under different experimental conditions. The selected labels investigated were (2-aminoethyl)trimethylammonium chloride hydrochloride (AETMA) as representative of the reductive amination mechanism and (hydrazinocarbonylmethyl) trimethylammonium chloride (Girard’s reagent T) as representative of hydrazone formation labeling chemistry. Purified samples were analyzed by CE with conductivity detection. A comparison of the purification selectivity and efficiency using the tested SPE materials is discussed in detail.
