Caracterização molecular e bioquímica de novas lipases obtidas por prospecção metagenômica com aplicação em biocatálise

Resumo: Esta tese reporta o trabalho realizado com duas lipases obtidas de uma biblioteca metagenômica, isto é, a clonagem, expressão, purificação e caracterização de uma nova lipase denominada LipC6G9 e a cristalização e determinação da estrutura cristalográfica da lipase LipC12. O gene da lipase LipC6G9 (lipC6G9) foi localizado juntamente com o gene de sua foldase, lifC6G9. LipC6G9 e LifC6G9 apresentam 96% e 77% de identidade, respectivamente, com as proteínas correspondentes de Aeromonas veronii B565. LipC6G9 e LifC6G9 foram coexpressas, ambas na forma N-truncadas, em Escherichia coli BL21 (DE3) usando os vetores pET28a(+) e pT7-7, respectivamente, e então purificadas por cromatografia de afinidade em coluna de Ni2+ HisTrap™ Chelating HP. A enzima purificada foi eluída da coluna complexada com a sua foldase (Lip-LifC6G9). As características bioquímicas e cinéticas deste complexo foram investigadas. As massas moleculares das proteínas N-truncadas são 32 kDa para LipC6G9 (incluindo a cauda com seis resíduos His) e 23 kDa para sua LifC6G9. Lip- LifC6G9 foi ativa em uma ampla faixa de pH (6,5-10,5) e temperatura (10-40 °C), sendo a maior atividade (1500 U mg-1) apresentada em pH 7,5 e 30 °C. A análise filogenética de LipC6G9 classificou-a como pertencente à subfamília I.1. A análise da seqüência com outras lipases bacterianas do Protein Data Bank mostraram que LipC6G9 apresenta a tríade catalítica (Ser99, Asp246, His268) com o pentapeptídeo conservado Gly-His-Ser-His-Gly (onde Ser é a serina catalítica), uma ponte dissulfeto e um sítio de ligação do íon cálcio conservados. Lip-LifC6G9 possui altas atividades específicas contra os substratos tributirina, tricaprilina e trioleína (1852, 1566 e 817 U mg-1, respectivamente). Estudos de imobilização do complexo no suporte hidrofóbico Accurel MP-1000 mostraram que a melhor concentração de proteína foi 5 mg/g (proteína/ suporte), com tempo de contato de 24 h, com uma eficiência de 68%. A atividade de hidrólise contra trioleína em nheptano para o preparado imobilizado foi 219 ± 28 U g-1, obtendo-se um aumento de 248% após imobilização. Quando Lip-LifC6G9 imobilizada foi incubada a 30 ºC em solventes orgânicos puros apresentou-se mais estável em n-heptano e em isopropanol. A imobilização não conduziu a um aumento da estabilidade térmica relativamente à enzima livre. Em reações de esterificação com ácido oléico e etanol, em presença de cossolventes (n-hexano e n-heptano), Lip-LifC6G9 imobilizada apresentou uma conversão de 95% em 6 h, com uma atividade de esterificação de 36 U g-1. As altas atividade e estabilidade de Lip-LifC6G9 livre e imobilizada sugerem que ela tem um bom potencial para uso em biocatálise e em reações de síntese orgânica. No caso da elucidação da estrutura tridimensional de LipC12, que também pertence a subfamília I.1, o cristal coletado com 2,64 Å de resolução pertence ao grupo espacial tetragonal, com parâmetros de cela a = b = 58,62, c = 192,60 Å e valores de R = 19,6% e Rfree = 25,4%. Esta é a primeira vez que a estrutura de uma lipase obtida pela abordagem metagenômica foi determinada. Este conhecimento proverá a base para aplicação de técnicas de engenharia genética, que podem viabilizar a obtenção e modificação LipC12 com características melhoradas para o uso em biocatálise. Abstract: This thesis reports on work undertaken with two lipases obtained from a metagenomic library, namely the cloning, purification and characterization of a new lipase denominated LipC6G9 and the determination of the crystallographic structure of lipase LipC12. Within the library, the gene lipC6G9 was cloned jointly with the gene for the foldase, lifC6G9. LipC6G9 and LifC6G9 have 96% e 77% identity, respectively, with the corresponding proteins of Aeromonas veronii B565. LipC6G9 and LifC6G9 were co-expressed, both in N-truncated form, in Escherichia coli BL21 (DE3) using the vectors pET28a(+) and pT7-7, respectively, and then purified by affinity chromatography using an Ni2+ column HisTrap™ Chelating HP. The purified enzyme eluted from the column complexed with the foldase (Lip-LifC6G9). The biochemical and kinetic characteristics of this complex were investigated. The molecular masses of the N-truncated proteins were 32 kDa for LipC6G9 (including the N-terminal His-tag with 6 residues) and 23 kDa for Lifc6G9. Lip-LifC6G9 was active over a wide range of pH values (6.5-10.5) and temperatures (10-40 °C), with the highest activity (1500 U mg-1) being obtained at pH 7.5 and 30 °C. A phylogenetic analysis placed LipC6G9 in the lipase subfamily I.1. Comparison of the sequence of LipC6G9 with those of other bacterial lipases in the Protein Data Bank showed that LipC6G9 contains the classic catalytic triad (Ser99, Asp246, His268) with the catalytic Ser occurring within a conserved pentapeptide (Gly-His-Ser-His-Gly), a conserved dissulfide bridge and a conserved calcium binding site. Lip-LifC6G9 has high specific activities against tributyrin, tricaprylin and triolein (1852, 1566 and 817 U mg-1, respectively). Studies of the immobilization of the complex on the hydrophobic support Accurel MP-1000 showed that the best protein concentration was 5 mg/g (protein/support), with a contact time of 24 h, with an efficiency of immobilization of 68%. The hydrolytic activity against triolein in n-heptane of the immobilized preparation was 219 ± 28 U g-1, 248% higher than that obtained before immobilization. When immobilized Lip-LifC6G9 was incubated at 30°C in pure organic solvents, it was most stable in n-heptane and in isopropanol. Immobilization did not lead to an increase in thermal stability relative to the free enzyme. For the esterification of oleic acid with ethanol, in the presence of co-solvent (n-hexane or n-heptane), immobilized Lip-LifC6G9 gave a conversion of 95% in 6 h, with a specific esterification activity of 36 U g-1. The high activity and stability of free and immobilized Lip-LifC6G9 suggest that it has a good potential for use in biocatalysis and organic synthesis reactions. In the case of the elucidation of the three dimensional structure of LipC12, which also belongs to lipase subfamily I.1, com 46% de identidade com a lipase de Pseudomonas aeruginosa PAL, the crystal collected to 2.70 Å resolution belongs the tetragonal space group P4122, with unit cell parameters a = b = 58.62, c = 192.60 Å, values of R = 19.6% e Rfree = 25.4%. This is the first time that the structure of a lipase obtained through a metagenomic approach has been determined. This knowledge provides the basis for the application of protein engineering techniques aimed at obtaining a modified LipC12 with improved characteristics for use in biocatalysis.
