Özet
Özet: Kanser hücreleri düzensiz, hızlı ve devamlı
bölünmeleri ve çoğalmaları açısından normal
hücrelerden farklıdırlar. Bu yüzden kanser hücreleri
oluştukları ilk yerde hızlıca bir tümör dokusu
oluşturmaya başlarlar. Tümör dokusu oluşup
büyüdükçe, zamanla dokudaki oksijen miktarı
azalmaya başlar. Oksijen miktarının azalmasıyla
tümör dokuda hipoksik bir ortam oluşur ve bu
hipoksik ortam gen ifadesinde değişikliklere neden
olur. Bu değişikliklerin oluşmasında dimerik
yapıda bulunan HIF-1 (Hipoksiyle indüklenebilen
faktör ) protein faktörlerinin önemli etkisi vardır.
Normalde vücutta hipoksik bir ortam oluştuğunda,
HIF-1 protein faktörleri gen ifadesini hipoksik
ortama yeni damarlar oluşturması(anjiyogenez)
yönünde değiştirir. Hipoksik ortam, oluşturulan damarlarla normoksik duruma geldiğinde ise
HIF-1 protein faktörleri protosomal yıkıma uğrarlar.
Tümörlü dokuda kanser hücrelerinin besin
ihtiyaçların artmasıyla ve dokuda hipoksik ortam
oluşmasıyla HIF-1 protein faktörleri tümörlü dokuya
anjiogenez oluşturulması yönünde genleri
regüle eder. Fakat, kanser hücrelerinin düzensiz,
hızlı ve devamlı büyümeleri, tümör dokunun sürekli
hipoksik kalmasına neden olur böylece HIF-1
protein faktörleri sürekli genleri anjiogenez oluşturmaları
yönünde regüle eder. Buna ek olarak
yapılan araştırmalara göre HIF-1 protein faktörlerinin
yıkımı için oksijene ihtiyaç vardır. Bu yüzden
hem hipoksik ortamın HIF-1 protein faktörlerini
tümör anjiyogenezi oluşturması yönünde uyarmasıyla,
hem de HIF-1 protein faktörlerinin proteozamal
yıkımında oksijene ihtiyaç duymasıyla,
bu çalışmada HIF-1 protein faktörlerinin anjiogenez
oluşturma mekanizmaları hakkında ve HIF-1
protein faktörlerini proteozamal yıkıma götüren
mekanizmaları hakkında araştırmalar geniş bir
perspektifle ele alınmıştır.
Abstract: Cancer cells differ from normal cells in terms of unregulated, rapid and continuous segregation and reproduction. Therefore, cancer cells can create tumor tissue in time where it first occur. As the tumor tissue grows, the amount of oxygen in the tissue starts to decrease. A hypoxic environment started to form in the tissue by lowering the amount of oxygen and this hypoxic environment effects the regulation of gene expression. Hypoxia Inducible Factor (HIF)-1 is a dimeric protein factor that mainly have role at gene expressions in hypoxia. Normally, when the body encounters a hypoxic environment, HIF-1 protein factors regulate the genes to create new vessels(angiogenesis) to the hypoxic tissue for supplying oxygen and nutrients and when the tissue turn to the its normal condition, these factors exposed proteasomal degradation. At a tumor tissue, due to the hypoxic environment, mainly because of rapid reproduction of cancer cells oxygen and nutrients are constantly needed by the cancer cells. When there is not enough oxygen, hypoxic environment stays permanent at the tumor tissue. HIF-1 protein factors regulate the genes that enhance the angiogenesis for cancer cells. Permanent hypoxic environment causes inducing of the HIF-1 protein factors to create tumoral angiogenesis continuously. In addition, according to the proved researches, oxygen is needed at the proteasomal degradation of HIF-1 protein factors. Therefore, because of both hypoxic environment induces HIF-1 protein factors to create tumoral angiogenesis and oxygen is needed to degradation of HIF-1 protein factors, at this review we want bring a clear view about the angiogenesis and its functional demands by emphasizing HIF-1 protein mechanism. We also mentioned about mechanism of gene regulation and mechanism of proteasomal degradation of HIF-1 protein factors to clarify the effects of the hypoxic environment.
