Resumo: Problemas econômicos, geopolíticos e ambientais relacionados ao petróleo e outros combustíveis fósseis levaram a um interesse crescente pelo desenvolvimento da produção de forma renovável de combustíveis e químicos de importância industrial. O n-butanol (C4H9OH), utilizado em larga escala na indústria, tem o potencial de ser produzido a partir de biomassa lignocelulósica por bactérias do gênero Clostridium, na chamada fermentação ABE (acetona, n-butanol e etanol). Muitos estudos se dedicam à otimização desse processo fermentativo e várias estratégias são empregadas a fim de obter altas concentrações de solventes. Tais estratégias, porém, podem se beneficiar de um conhecimento aprofundado do metabolismo do microrganismo. Desta forma, o presente trabalho teve como principal objetivo a analise do transcriptoma e do perfil fermentativo da bactéria Clostridium saccharoperbutylacetonicum DSM14923 durante seu crescimento e fermentação em hidrolisado lignocelulósico de cana-de-açúcar suplementado com polpa celulósica e complexo enzimático de celulases e hemicelulases, para compreender o metabolismo do microrganismo em condição favorável para uma alta produção de solventes. Os dados obtidos pela analise da transcrição gênica, aliados aos resultados do perfil fermentativo, permitiram um melhor entendimento quanto a pontos-chave do metabolismo fermentativo, a obtenção de informações novas quanto ao funcionamento da regulação gênica e a sugestão de possíveis formas de melhorar linhagens de Clostridium para a produção de solventes

Abstract: Environmental, geopolitical and economic problems related to oil and other fossil fuels led to an increased interest for the development of renewable production of industrially important fuels and chemicals. N-butanol (C4H9OH), a chemical of extensive use in the industry, has the potential to be produced from lignocellulosic biomass by bacteria from the genus Clostridium, through the so-called ABE fermentation (acetone, n-butanol and ethanol). Several studies are dedicated to the optimization of this fermentation process and several strategies are used to yield high solvent concentrations. These strategies, however, can benefit from in-depth knowledge on the microorganisms metabolism. Therefore, this study had as main objective the analysis of the transcriptome and fermentative profile from the bacteria Clostridium saccharoperbutylacetonicum DSM14923 during its growth in sugar-cane lignocellulosic hydrolysate supplemented with cellulosic pulp and cellulases and hemicellulases enzymatic complex, to comprehend the microorganisms metabolism in a favorable condition for high solvents production. The data obtained from gene transcription analysis, together with the fermentative profile results, allowed for a better understanding regarding fermentative metabolism key-elements, the acquirement of new information regarding gene regulation mechanisms and the suggestion of possible ways to further improve Clostridium strains for solvent production
