This PhD thesis was performed in the laboratory of Professor Markó, at the Chemistry Department of the Université catholique de Louvain.
Our research concerned the synthesis of Amphidinol 3, a natural product recently isolated from different dinoflagelates from the Asian sees. This compound showing a complex structural framework displayed unprecedented biological activities. Nevertheless, due to the small amount of product available in the natural source, no medicinal application can be envisaged and synthesis is the only reliable approach for large scale treatment development.
In addition to our efforts for the construction of this complex molecule, the developed methodologies proved to be applicable in a large scope of applications. Different novel approaches to polyenic functions were developed and we now dispose of a short and efficient strategy for the construction of trienes. Applied to the Amphidinol 3, this strategy proved to be the most efficient among all the published approaches so far. We next prepared the highly functionalized tetrahydropyranes of our target molecule by novel methodologies.
