Abstract
Not much is known to date about the candidate division OP3. A double density gradient centrifugation of a methanogenic enrichment led to a fraction containing over 80 % OP3-cells. After the extraction of genomic DNA 5 contigs with an overall length of 1.65 Mbp were obtained via sequencing and assigned to OP3. An automatic annotation gave a first insight into the genome of candidate division OP3 revealing a NADH-hydrogenase and a D-lactate-dehydrogenase. Electron micrographs of these enrichment cultures showed single OP3-cells on the filter and attached to larger rod-shaped cells. The hypothesis that OP3 grows on formate as substrate was disproven In addition OP3 was not able to utilize hydrogen, succinate, acetate, propionate and butyrate as energy and carbon source. The next step would be a physiological characterization of OP3 based on the information provided by the partial genome.
